The monograph discusses models of synthetic protocells, which are cell-like structures obtained from non-living matter endowed with some rudimentary kind of metabolism and genetics, but much simpler than biological cells. They should grow and proliferate, generating offsprings that resemble in some way the parent protocells with some variation, so that selection may take place. Sustainable protocell populations have not yet been obtained experimentally and mathematical models are therefore extremely important to address key questions concerning their synthesis and behavior. Different protocell “architectures” have been proposed and high-level abstract models like those that are presented in this book are particularly relevant to gain a better understanding of the different properites. These models are able to treat all the major dynamical phenomena in a unified framework, so they can be seen as “virtual laboratories” for protocell research. Particular attention is paid to the problem of synchronization of the fission rate of the whole protocell and the duplication rate of its "protogenetic" material, which is shown to be an emergent property that spontaneously develops in successive generations. The book is of interest for a broad range of scientists working in soft matter physics, chemistry and biology, interested in the role protocells may play on the development of new technologies with medical, environmental and industrial applications as well as scientists interested in the origin of life.

This interdisciplinary book consists of the proceedings of the Alexander Ivanovich Oparin lOOth Anniversary Conference, The Third Trieste Conference on Chemical Evolution, which took place at the International Centre for Theoretical Physics from 29 August till 2 September, 1994. A general overview of Oparin's life and work is followed by a review of Alfonso Herera, another pioneer in the studies of the origin of life. The subject matter is organized in ten sections corresponding to various aspects of our current understanding of the subject that was initiated by Oparin. These subjects were covered by fifty three speakers. There were sixty seven participants from a wide geographical distribution; twenty seven countries were represented. In this study, various protocell models have been constructed de novo with the bottom-up approach. Here we describe a silica-based protocell composed of a nanoporous amorphous silica core encapsulated within a lipid bilayer built by self-assembly that provides for independent definition of cell interior and the surface membrane. In this review, we will first describe the essential features of this architecture and then summarize the current development of silica-based protocells at both micro- and nanoscale with diverse functionalities. As the structure of the silica is relatively static, silica-core protocells do not have the ability to change shape, but their interior structure provides a highly crowded and, in some cases, authentic scaffold upon which biomolecular components and systems could be reconstituted. In basic research, the larger protocells based on precise silica replicas of cells could be developed into geometrically realistic bioreactor platforms to enable cellular functions like coupled biochemical reactions, while in translational research smaller protocells based on mesoporous silica nanoparticles are being developed for targeted nanomedicine. Ultimately we see two different motivations for protocell research and development: (1) to emulate life in order to understand it; and (2) to use biomimicry to engineer desired cellular interactions.

The lipid bilayer can be modified with targeting ligands or other ligands to achieve targeted delivery of cargo components that are loaded within the protocell to a target cell, e.g., a type of cancer. Shielding materials can be conjugated to the surface of the lipid bilayer to reduce undesired non-specific binding.
